The short answer
Tirzepatide and retatrutide are incretin receptor agonists: tirzepatide binds the GIP and GLP-1 receptors, and retatrutide - listed in this catalog as GLP-3 (RT) - adds the glucagon receptor as a third target. Cagrilintide is an analogue of amylin, a hormone co-secreted with insulin from pancreatic beta cells. Amylin acts through amylin receptors, which are built from the calcitonin receptor paired with accessory proteins. That is a different receptor system from the incretin receptors the other two engage.
| Cagrilintide | Tirzepatide | Retatrutide | |
|---|---|---|---|
| Hormone it mimics | Amylin | GIP and GLP-1 | GIP, GLP-1 and glucagon |
| Receptors engaged | Amylin receptors (calcitonin receptor based) | GIP and GLP-1 receptors | GIP, GLP-1 and glucagon receptors |
| Design feature | Acylated for a long half-life | Single molecule, two receptor activities | Single molecule, three receptor activities |
| Published human data | Phase 2, plus combination studies | Large late-stage trial programme | Phase 2; phase 3 programme under way |
Two hormone systems, three molecules.
Cagrilintide: an amylin analogue
Native amylin is short-lived, which limits its use as a research tool. Cagrilintide was developed as a long-acting, acylated amylin analogue so that its activity persists. Published mechanistic work describes it acting through brain amylin receptors, specifically the AMY1 and AMY3 subtypes, rather than through any incretin receptor.
Much of the published human research studies cagrilintide alongside semaglutide rather than on its own, because the two act on separate receptor systems. That design choice is itself a statement about mechanism: researchers pair them precisely because they do not overlap.
Tirzepatide and retatrutide: the incretin lineage
Tirzepatide and retatrutide differ from each other by receptor count. Tirzepatide is a dual agonist of the GIP and GLP-1 receptors. Retatrutide adds activity at the glucagon receptor, making it a triple agonist. Our comparison of semaglutide, tirzepatide and retatrutide covers that lineage in more detail.
Evidence quality
Tirzepatide has the largest published human trial record of the three, from a late-stage clinical programme. Retatrutide and cagrilintide both have published phase 2 data, and cagrilintide additionally has published studies in combination with semaglutide. For a research design, the practical difference is how much comparable published data exists for each molecule, not a ranking of the molecules themselves.
Handling notes
All three are supplied as lyophilized powders and are handled like other peptide reference materials: stored cold and dry before reconstitution, and protected from repeated freeze-thaw afterwards.
Compounds referenced here are supplied as analytical-grade reference materials for in-vitro and laboratory research only. Nothing on this page is a recommendation for human or veterinary use.
Frequently asked questions
What is the difference between cagrilintide and retatrutide?
Cagrilintide is a long-acting analogue of amylin and acts through amylin receptors. Retatrutide is a triple incretin-family agonist acting on the GIP, GLP-1 and glucagon receptors. They work through different hormone systems.
Is cagrilintide a GLP-1 agonist?
No. Cagrilintide is an amylin analogue. It acts through amylin receptors, which are built from the calcitonin receptor and accessory proteins, not through the GLP-1 receptor.
Why is cagrilintide often studied with semaglutide?
Cagrilintide and semaglutide act on separate receptor systems, amylin and GLP-1 respectively. Published combination studies pair them because their mechanisms do not overlap.
Which of the three has the most published human research?
Tirzepatide has the largest published human trial record of the three. Retatrutide and cagrilintide have published phase 2 data, and cagrilintide also has published combination studies with semaglutide.
