Why Is My Reconstituted Peptide Solution Cloudy?

A cloudy vial after adding diluent is one of the most common things a researcher notices, and it is usually not what it looks like. There are four distinct causes, they look similar in the first minute, and they diverge quickly. This page separates them.

The short answer

Most cloudiness seen immediately after adding diluent is undissolved material, not damaged material. A lyophilized cake does not dissolve instantly, and a solution that is hazy at thirty seconds is frequently clear at ten minutes with no intervention at all. Cloudiness that persists after the material has had time to dissolve is a different signal and is worth taking seriously.

1. It simply has not dissolved yet

A lyophilized peptide is a dry, porous cake. Diluent has to penetrate it, and that takes time - particularly for larger vial masses and for peptides that are less readily soluble. Adding diluent down the side of the vial and then leaving it undisturbed for five to fifteen minutes resolves the majority of cloudiness reports without any further action.

2. It was shaken

Peptides are chains that hold a shape, and mechanical agitation can drive them to aggregate with one another. Vigorous shaking introduces air, creates foam, and can produce a persistent cloudiness that gentle handling would not have. This is the one cause that is directly created by the handler, and it is not reversible by further shaking.

The standard technique is to add diluent slowly against the vial wall rather than directly onto the cake, then roll the vial between the fingers or swirl it gently. Never shake, and never vortex unless a protocol specifically calls for it.

3. It is precipitating

Some peptides have limited solubility at higher concentrations, and a solution that would be clear at one concentration can be cloudy at another. Choosing a lower concentration - more diluent for the same vial mass - is the usual response. Diluent choice matters too: bacteriostatic water is the standard for a reason, and substituting something else changes the chemistry the material was characterised in.

4. It is contaminated

Contamination is the least likely cause on day one and the most likely cause on day five. A solution that was clear and has become cloudy in storage, particularly with visible material or a changed smell, should be discarded rather than investigated. Bacteriostatic water suppresses microbial growth but does not sterilise, and a vial entered many times with a used needle is the common route in.

What to do, in order

Does cloudiness mean the peptide was low purity?

Cloudiness is a handling and solubility observation, not a purity measurement. Purity is established by HPLC on the lot and reported on the Certificate of Analysis. A hazy solution from a high-purity lot is far more often a dissolution or concentration issue than a purity one.
